1. Why Taq amplified PCR product cannot be cloned by blunt end ligation method??

a.       In general the both blunt end and sticky end are common cloning method. Blunt ends are produced when the DNA are cut at same point with no protruding ends. In case of sticky ends there will be 2 protruding ends (unpaired region or over hangs). DNA digestion with certain set of enzymes produce sticky ends where as with other enzymes produces blunt ends.
Taq amplified PCR products cannot be cloned by blunt end ligation method because it produces adenine over hangs in their 3’ ends. Hence it will  not be suitable for sticky end cloning method. Therefore TA cloning is the common method used for cloning PCR amplified products.
